A recombinant rabbit monoclonal reagent from Antibodies.com targeting the replication-independent histone variant H3.3, supplied as an unconjugated, BSA-stabilized liquid in TBS (pH 7.4) with 40% glycerol and 0.05% sodium azide. The clone is validated for western blot, immunocytochemistry/immunofluorescence, immunohistochemistry, and chromatin immunoprecipitation on human, mouse, and rat samples, supporting studies of transcriptionally active chromatin, telomeric nucleosomes, and promoter/enhancer occupancy.

Histone H3.3 differs from canonical H3.1/H3.2 by only a handful of residues (positions 31, 87, 89, and 90), yet marks nucleosomes deposited outside S-phase across gene bodies, regulatory elements, and pericentromeric regions. Researchers use H3.3 immunolabeling to map epigenetic remodeling during differentiation, reprogramming, and development, and to detect recurrent oncohistone substitutions (K27M, G34W, G34R/V) characteristic of pediatric high-grade gliomas and giant cell tumors of bone. ChIP-grade specificity is compatible with both native and cross-linked chromatin workflows.

Each vial contains 100 µl of antibody. Store at -20°C; aliquot on first use to minimize freeze-thaw cycles, and protect from light during extended bench handling. For FFPE tissue sections, heat-induced antigen retrieval with citrate buffer (pH 6.0) is recommended. The unconjugated format allows flexible pairing with HRP, biotin, or fluorophore-conjugated secondary reagents.
