HINT1 (Histidine Triad Nucleotide-Binding Protein 1) is a 126-amino-acid homodimeric purine phosphoramidase expressed in the cytoplasm and nucleus of human, mouse, and rat tissues. This unconjugated mouse monoclonal IgG reagent from Antibodies.com, assigned clone #A3912, detects the ~13-14 kDa target for western blot, immunohistochemistry, immunofluorescence, and flow cytometry workflows. Because the antibody is supplied without a fluorophore or enzyme label, researchers can pair it with HRP-, biotin-, or fluorophore-conjugated secondaries matched to their detection platform.

Functionally, HINT1 acts as a haploinsufficient tumor suppressor whose catalytic His110/His112/His114 triad hydrolyzes purine nucleotide phosphoramidate substrates, including AMP-morpholidate and AMP-N-alanine methyl ester intermediates relevant to phosphoramidate prodrug activation. The protein is recruited to ionizing radiation-induced foci, where it supports γ-H2AX acetylation and ATM checkpoint signaling during the DNA damage response, and it modulates apoptosis through p53, Bax, and Bcl-2 regulation. Downregulation of HINT1 has been documented in hepatocellular carcinoma, colon cancer, gastric tumors, osteosarcoma, MCF-7 and HeLa lines, while biallelic HINT1 mutations in humans are linked to axonal neuropathy and neuromyotonia, making the protein a target of interest in both oncology and neuroscience research.

Each vial contains 100 µl of liquid antibody in PBS-based buffer. Shipped cold; aliquot upon arrival and store at -20 °C, avoiding repeated freeze-thaw cycles to preserve activity. For research use only.
