The HIF1A polyclonal antibody is raised in rabbit and supplied by Antibodies.com as an unconjugated reagent in a 100 µl vial, ready for use in western blot and immunohistochemistry workflows. HIF1A (Hypoxia-Inducible Factor 1-Alpha) is the oxygen-regulated subunit of the HIF-1 transcription factor, normally hydroxylated and targeted for proteasomal degradation under normoxia, but stabilized and translocated to the nucleus when cellular oxygen falls. Detecting endogenous HIF1A therefore requires attention to oxygen tension during sample handling: many users culture or treat cells under hypoxic conditions, or use chemical stabilizers such as cobalt chloride or desferrioxamine, before lysis to ensure the protein is detectable, and compare against normoxic controls.

This reagent was validated for reactivity against human, mouse, and rat samples, making it useful across species in studies of tumor angiogenesis, ischemic injury, metabolic adaptation, and other hypoxia-related pathways. For western blot, whole-cell lysates are typically resolved by SDS-PAGE and probed at a dilution appropriate for chemiluminescent or fluorescent detection, with the expected band appearing near 93 kDa depending on post-translational modification. For IHC, antigen retrieval and peroxidase- or fluorescence-based detection systems are commonly paired with this polyclonal reagent to localize HIF1A staining in formalin-fixed tissue sections. Aliquot upon receipt and store refrigerated to preserve activity across multiple freeze-thaw cycles.
