Generated in rabbit and supplied by Antibodies.com as an unconjugated polyclonal antiserum, this reagent recognizes the hypoxia-inducible factor 1-alpha (HIF-1α) protein, the oxygen-regulated subunit of the HIF-1 transcription factor. Under normoxia, conserved proline residues in HIF-1α are hydroxylated by prolyl hydroxylase domain (PHD) enzymes, leading to von Hippel-Lindau (VHL)-mediated ubiquitination and proteasomal clearance. Under hypoxia, iron chelation, or pathological pseudohypoxic signaling, the protein is stabilized, dimerizes with HIF-1β, and translocates to the nucleus where the complex activates transcription of genes controlling glycolysis, angiogenesis, erythropoiesis, and cell survival decisions.

The antibody is provided at 100 µl of liquid stock and is validated for western blot (WB), immunohistochemistry (IHC), and ELISA across human, mouse, and rat samples. On western blots, the HIF-1α band appears near 120 kDa, consistent with the full-length isoform. Because the protein pool turns over rapidly with changing oxygen conditions, lysates are usually prepared with protease and phosphatase inhibitor cocktails and handled cold to preserve labile HIF-1α. In tumor and ischemic tissue sections, immunostaining is widely used as a marker of regional hypoxia and as a downstream readout of pathways such as PI3K/AKT/mTOR, and it supports research into cancer progression, cardiovascular ischemia, and inflammatory hypoxia responses.
