Hematopoietic Cell Kinase (HCK) is a Src-family non-receptor tyrosine kinase expressed primarily in myeloid cells and B lymphocytes, where it controls phagocytosis, integrin signaling, and inflammatory responses. The polyclonal reagent from Antibodies.com is raised in rabbit and affinity-purified against a synthetic phosphopeptide spanning tyrosine 522 in the C-terminal negative-regulatory tail of human HCK. Phosphorylation at Y522 stabilizes the closed, autoinhibited conformation of the kinase, so recognition of pY522 is used as a marker for the regulated, low-activity state of the enzyme in primary immune cells and in leukemic cell lines.

The antibody is supplied as 100 µl of unconjugated IgG in a vial and is validated for Western blot on human samples. Bands are typically detected in the ~59–61 kDa region, matching the known p59 and p61 HCK isoforms. Researchers studying myeloid differentiation, Fc-receptor signaling, or B-cell receptor crosstalk can use this reagent to monitor dynamic changes in HCK Y522 phosphorylation downstream of cytokine, integrin, or pharmacological kinase inputs.

For best results, storage at -20 °C is recommended, with the antibody dispensed into small aliquots to limit freeze-thaw cycles and protected from light. The product is intended for research use only and is not validated for diagnostic or therapeutic applications.
