A polyclonal antibody reagent from Antibodies.com raised in rabbit against HAVCR2 (also known as TIM-3), a type I transmembrane immune checkpoint receptor expressed on interferon-gamma-producing T cells, regulatory T cells, macrophages, and dendritic cells. TIM-3 acts as a co-inhibitory molecule that contributes to T cell exhaustion in chronic viral infection and in the tumor microenvironment, and it is an active focus of immuno-oncology research alongside PD-1 and LAG-3.
This unconjugated reagent is provided at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, shipped as a 100 µl vial that can be diluted into standard immunodetection buffers. Validated uses include Western blot detection of HAVCR2 in cell or tissue lysates and immunohistochemistry on fixed tissue sections. Cross-reactivity with both human and mouse HAVCR2 allows parallel profiling of patient specimens and corresponding mouse models, supporting translational studies of checkpoint biology.
Working concentrations depend on the detection method and antigen abundance, and end users typically optimize the dilution empirically against a positive-control lysate or tissue. For long-term use, store the vial at -20 °C; the 50% glycerol cryoprotectant limits damage from freeze-thaw cycles and brief benchtop handling. Because sodium azide is included as a preservative, downstream enzymatic reactions, particularly horseradish peroxidase-based detection, require thorough washing to remove residual inhibitor.
Common research applications include profiling TIM-3 expression on tumor-infiltrating lymphocytes, characterizing exhaustion phenotypes in chronic viral infection models, and screening candidate TIM-3 ligands such as galectin-9 and CEACAM1. The reagent also supports immunohistochemistry studies of autoimmune tissue and time-course Western blots tracking checkpoint modulation after drug treatment.
