HAO1 (hydroxyacid oxidase 1) is a peroxisomal FMN-dependent enzyme most highly expressed in hepatocytes and proximal renal tubule cells, where it catalyzes the oxidation of glycolate to glyoxylate. Hepatic flux through this enzyme is the major precursor of endogenous oxalate, the metabolite that accumulates in primary hyperoxaluria, and pharmacologic inhibition of HAO1 is being pursued as a strategy to lower oxalate production in primary and enteric hyperoxaluria. A specific antibody reagent is therefore useful for expression studies, target validation, and biomarker work in this disease area.

This rabbit polyclonal antibody from Antibodies.com is raised against HAO1 and supplied as 100 µl of a 1 mg/ml stock in PBS with 0.1% sodium azide and 50% glycerol. The reagent is unconjugated, making it compatible with indirect detection using HRP- or fluorophore-conjugated secondary antibodies, and has been validated for western blot and immunohistochemistry, with reactivity confirmed against human and mouse samples. The polyclonal format generates broad epitope coverage, useful when mapping expression across tissues or when single-epitope monoclonal reagents give weak signal.

For best stability, aliquot upon receipt to limit freeze-thaw cycles and store at -20°C in the supplied glycerol buffer; the azide-containing formulation also allows short-term use at 4°C. Researchers using this reagent on liver and kidney sections can expect cytoplasmic and peroxisomal staining reflecting the known subcellular distribution of HAO1, and the unconjugated format gives flexibility to pair it with a custom secondary antibody for multiplexed immunofluorescence or tyramide signal amplification protocols.
