GCAP1, the ~23 kDa calcium-binding photoreceptor protein encoded by GUCA1A, governs Ca2+-sensitive regulation of retinal guanylyl cyclase 1 (RetGC1) during cGMP replenishment following photobleaching. The protein concentrates in cone outer and inner segments and, to a lesser extent, in rod inner and outer segments, where it switches from a guanylyl cyclase activator at low free calcium to an inhibitor as Ca2+ returns to its dark resting level.

This unconjugated rabbit polyclonal IgG reagent from Antibodies.com is supplied as a 100 µl vial of polyclonal antiserum validated for western blot and immunohistochemistry on mouse-derived samples, supporting immunodetection of GCAP1 in retinal tissue. The antibody is useful for inherited retinal disease research, including autosomal dominant cone dystrophy (COD3) and cone-rod dystrophy (CORD14), in which GUCA1A mutations such as Y99C, P50L, L151F, and E111V produce constitutively active RetGC1, elevated outer-segment calcium, and progressive photoreceptor cell loss. Characterized knock-in (E111V) and transgenic (Y99C, L151F) mouse models have used comparable polyclonal reagents to map altered GCAP1 biodistribution, Mertk downregulation, and synaptic and inflammatory changes across the outer nuclear layer, and wild-type GCAP1 supplementation studies are exploring biochemical rescue of these photoresponse defects.

The reagent should be stored refrigerated and aliquoted upon first use to limit repeated freeze-thaw cycles and preserve binding activity across immunoblot and immunostaining experiments.
