The polyclonal reagent offered by Antibodies.com is raised in rabbit against GRIN1, the gene that encodes the NR1 (GluN1) subunit of the N-methyl-D-aspartate (NMDA) receptor. NR1 is the obligatory subunit of every functional NMDA receptor, a ligand-gated ion channel that mediates calcium-permeable excitatory signaling at central nervous system synapses. This unconjugated primary antibody is supplied as a 100 µL aliquot and reacts with human GRIN1, supporting work on excitatory neurotransmission, synaptic plasticity, and excitotoxic mechanisms in neuronal systems.

Validated for western blotting (WB) and ELISA workflows, the reagent can be used to detect endogenous NR1 protein in cell and tissue lysates as well as recombinant preparations from heterologous expression systems. The polyclonal format recognizes several epitopes across the NR1 subunit, which is useful when probing the different splice variants generated by alternative splicing at the N1 cassette and C2' cassette regions, including the long and short C-terminal isoforms. For storage and handling, the antibody should be kept refrigerated and protected from light, with aliquoting recommended after first use to limit freeze-thaw cycles and preserve binding activity.

Common research applications include studies of NMDA receptor trafficking, phosphorylation, surface expression, and involvement in neurological disease models ranging from ischemic injury and epilepsy to schizophrenia, Alzheimer's disease, and GRIN1-related neurodevelopmental disorders. Researchers investigating receptor assembly or subunit-specific pharmacology can pair this reagent with antibodies against GluN2 subunits or postsynaptic density markers to characterize NMDA receptor composition in their experimental system.
