From Antibodies.com, this rabbit polyclonal primary antibody targets endogenous Golgi phosphoprotein 3 (GOLPH3), the ~34 kDa peripheral membrane phosphoprotein that localises to the cytoplasmic face of the trans-Golgi network through its C-terminal PI(4)P-binding GPP34 domain. Also known by the synonyms GPP34, MIDAS, GOPP1 and coat protein GPP34, GOLPH3 maintains Golgi ribbon architecture, intra-Golgi vesicle trafficking, and retromer-mediated receptor recycling from endosomes. In cancer research it is widely studied because its overexpression constitutively activates both mTORC1 and mTORC2 signalling, and is recurrently amplified in solid tumour cohorts including breast, gastric, colorectal, lung and glioblastoma specimens where it correlates with poor prognosis and rapamycin sensitivity in xenograft models.
Supplied as a 200 µL liquid aliquot, the unconjugated rabbit polyclonal IgG reagent has been validated on human and mouse samples for Western blot, immunohistochemistry of fixed tissue sections and flow cytometry. Research applications include immunoblotting of cultured cell lysates to assess endogenous GOLPH3 levels, IHC staining of tumour and normal tissue microarrays, and intracellular flow cytometry staining where the antibody functions as a marker of trans-Golgi morphology. The reagent supports investigation of GOLPH3-driven cell migration, Golgi-mediated sorting and mTOR pathway crosstalk, and pairs readily with phospho-S6K or phospho-Akt antibodies to monitor downstream signalling in the same lysate or section. The liquid format allows direct dilution into standard assay buffers; intended for research use only.
