Anti-GGA2 Antibody

A rabbit polyclonal antibody against GGA2, an ARF-binding clathrin adaptor that shuttles cargo between the trans-Golgi network and endosomes, is supplied by Antibodies.com as 100 µl of unconjugated antiserum. The reagent detects the endogenous ~67 kDa GGA2 protein in human, mouse, and rat samples, and it has been validated for Western blot, immunohistochemistry, and immunofluorescence workflows.

GGA2 belongs to the GGA family of vesicular transport adaptors. Its VHS domain recognizes DXXLL-type sorting motifs on cargo such as the cation-independent mannose 6-phosphate receptor, while its GAT domain binds ARF-GTP and ubiquitin at the Golgi membrane. Through these interactions, GGA2 packages hydrolase receptors into clathrin-coated buds at the TGN, contributes to receptor recycling, and supports lysosomal enzyme delivery.

For Western blot, the antibody labels a single band near the predicted molecular weight in whole-cell lysates from HEK293, HeLa, and NIH-3T3 lines. On fixed cells for immunofluorescence, it produces a compact perinuclear pattern that colocalizes with TGN46, consistent with Golgi/TGN localization, and shows punctate endosomal staining after a brief serum chase. In IHC, it has been applied to rodent brain sections to map regional GGA2 expression.
