This rabbit polyclonal antibody from Antibodies.com is a phospho-specific reagent raised against GRB2-associated binding protein 1 (Gab 1) phosphorylated at tyrosine 627. It is supplied as 100 µl of a 1 mg/ml stock in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2.

Gab 1 functions as a scaffold and adaptor downstream of several receptor tyrosine kinases, including the HGF receptor Met, where phosphorylation of C-terminal tyrosines such as Y627 creates docking sites for SH2-domain effectors like SHP-2 and links growth-factor input to Ras/MAPK and PI3K/Akt signaling. Detecting the modified form reports on pathway activation rather than total Gab 1 abundance, making phospho-Y627 a useful marker in studies of cytokine, growth-factor, and integrin signalling.

Validated for Western blot on human and rat samples, the reagent is unconjugated and is therefore compatible with HRP- or fluorophore-conjugated secondary antibodies chosen by the end user. The format is a 100 µl vial at 1 mg/ml; aliquot on receipt to limit repeated freeze-thaw cycles, store short term at 4 °C, and hold longer term at -20 °C protected from light. The buffer contains 0.05% sodium azide as a preservative.

Because the host is rabbit, anti-rabbit IgG secondary reagents can be used for detection. The polyclonal bleed provides a mixed population of binders against the phospho-Y627 epitope of Gab 1, so a single reagent can detect the modified target in lysates from both human and rat origins.
