This recombinant rabbit monoclonal antibody from Antibodies.com is raised against a phosphorylated epitope on FOXO3A and specifically recognizes the serine 253 residue when it is phosphorylated by AKT/PKB signaling. Because S253 phosphorylation drives FOXO3A out of the nucleus and suppresses its transcriptional activity on genes involved in apoptosis, oxidative stress response, and cell cycle arrest, a phospho-selective reagent is the standard tool for tracking this regulatory switch in cell signaling studies.

The reagent is supplied as 100 µl of unconjugated antibody in a buffer of 1×TBS at pH 7.4 with 1% BSA, 40% glycerol, and 0.05% sodium azide as preservative. It is validated for western blot and immunohistochemistry applications on human, mouse, and rat samples, and as a clonally purified recombinant molecule produced in rabbit host cells it offers improved lot-to-lot consistency compared with traditional polyclonal sera for reproducible detection of the phospho-S253 mark in lysates and fixed tissue sections.

Suggested uses include western blot analysis of cell or tissue lysates to monitor AKT pathway activation, immunohistochemistry on formalin-fixed paraffin-embedded sections to localize phospho-S253 FOXO3A in tumor or neural tissue, and matched immunoblot and IHC comparison between treated and untreated samples to confirm pathway engagement under different experimental conditions.
