FOXO3 (Forkhead box O3) is a Forkhead-family transcription factor whose activity is gated by AKT/PKB-mediated phosphorylation, with serine 253 acting as one of the primary regulatory sites that drive 14-3-3 binding and cytoplasmic retention. This phospho-specific reagent from Antibodies.com recognizes the phosphorylated S253 epitope, letting researchers read out PI3K/AKT pathway activity and probe downstream effects on apoptosis induction, autophagy regulation, oxidative-stress responses, and cell-cycle arrest in cancer signaling and longevity studies.

Supplied as 100 µl of unconjugated rabbit polyclonal IgG, the antibody is validated for western blot, immunohistochemistry, and immunofluorescence across human, mouse, and rat samples. The unconjugated format and polyclonal host give flexibility in selecting secondary reagents and amplification chemistries. To preserve phospho-epitope integrity, aliquot the vial upon arrival and store at -20 °C, avoiding repeated freeze-thaw cycles.
