This polyclonal antibody from Antibodies.com specifically recognizes human and mouse FOXO1 when phosphorylated at the serine 256 residue, a regulatory site directly modified by Akt/PKB kinase signaling. FOXO1 (Forkhead box protein O1), originally identified as the rhabdomyosarcoma-associated fusion partner FKHR, is a forkhead-family transcription factor that controls genes involved in glucose metabolism, cell-cycle arrest, oxidative stress response, and insulin signaling. Phosphorylation at Ser256 drives FOXO1 export from the nucleus and its sequestration in the cytoplasm, attenuating transcription of pro-apoptotic and metabolic target genes downstream of PI3K/Akt.

Supplied as a 100 µl vial of unconjugated rabbit polyclonal IgG, the reagent is validated for Western blot detection of endogenous levels of phospho-FOXO1 in lysates derived from Human and Mouse samples. Researchers use this reagent to monitor PI3K/Akt pathway activity downstream of insulin and growth-factor receptors, characterize cancer cell lines and primary tumors with dysregulated PI3K-Akt signaling, track the response of metabolic tissues such as liver, skeletal muscle, and adipose depots to fasting and refeeding, and investigate oxidative-stress phenotypes in cells undergoing apoptosis or autophagy.
