This mouse monoclonal IgG2a antibody from Antibodies.com is raised against FOXO1 (forkhead box O1) and binds the endogenous protein in human and mouse samples for western blot and immunocytochemistry workflows.
FOXO1 is a forkhead-family transcription factor central to insulin and IGF-1 receptor signaling. AKT-mediated phosphorylation drives FOXO1 out of the nucleus, so its subcellular localization is widely used as a read-out of PI3K/AKT pathway activity. The protein also regulates hepatic gluconeogenesis, adipocyte differentiation, oxidative-stress responses, autophagy, and G1 cell-cycle arrest through targets such as p27/Kip1 and G6PC, and it is studied as a candidate tumor suppressor in prostate, breast, and several hematopoietic cancers.
The unconjugated reagent is supplied as a 100 µl liquid aliquot and is validated on cultured cells and tissue sections for western blot and immunocytochemistry. Its IgG2a framework pairs with standard anti-mouse secondary reagents, including HRP, biotin, Alexa Fluor, and DyLight conjugates. The clone works on formaldehyde- or methanol-fixed specimens and tolerates detergent permeabilization, making it usable on established cell lines and primary cells in parallel.
Typical experimental designs pair FOXO1 detection with phospho-AKT or total AKT staining, insulin or IGF-1 stimulation, and readouts of downstream targets such as p27 and G6PC. Nuclear versus cytoplasmic scoring using a DAPI or histone H3 co-stain is used to quantify pathway activity. The reagent also serves as a control in CRISPR and RNAi FOXO1-knockdown studies, where it confirms target loss alongside functional assays.
Storage and handling: keep refrigerated for short-term use; aliquot and freeze at -20 °C for longer storage; limit freeze-thaw cycles; protect from light when used with fluorescent secondary antibodies.
