Rabbit polyclonal IgG reagent from Antibodies.com raised against the Forkhead box A2 transcription factor (FOXA2), a winged-helix pioneer factor mapped to human chromosome 20p11.21 (UniProt Q9Y261, gene ID 3170) and synonymous with HNF-3β and TCF3B. Each vial contains 200 µL of unconjugated antiserum, affinity-purified from rabbits immunized with a synthetic peptide, and supplied in phosphate-buffered saline with 50% glycerol and 0.02% sodium azide. Aliquot and store at -20 °C to protect activity through repeated freeze-thaw handling.

Validated on endogenous FOXA2 from human and rat specimens in western blot, immunohistochemistry, and immunofluorescence workflows, the reagent consistently resolves a target band near 48 kDa by SDS-PAGE/WB, with the phosphorylated species migrating between 55-65 kDa in HepG2 and HeLa lysates. Useful starting dilutions sit at 1:500 to 1:1000 for western blot and 1:50 to 1:200 for IF/ICC, though titration on a representative lysate or section is recommended before scaling. Pair with an HRP- or Alexa Fluor-conjugated goat anti-rabbit IgG secondary for chromogenic, fluorescence, or near-infrared detection.

FOXA2 acts as a pioneer factor that opens compacted chromatin at lineage-specific enhancers, directing definitive endoderm differentiation, hepatic specification, pancreatic β-cell development via Pdx1 enhancer priming, and surfactant production in type II pneumocytes. The reagent is referenced in lung and gastric adenocarcinoma profiling, type-II-diabetes and metabolic disease models, hepatocyte lineage-tracing, and ChIP studies of the albumin and Pdx1 enhancers in foregut-derived lineages. For research use only; not validated for diagnostic or therapeutic applications.
