This polyclonal antibody from Antibodies.com specifically recognizes FKHR (also known as FOXO1A) when phosphorylated at serine 256, a regulatory site modified by Akt/PKB kinase downstream of insulin and growth factor signaling. Raised in rabbit, the reagent is supplied at 1 mg/ml in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2, in a 100 µl vial.

It is validated for Western blot and immunohistochemistry (IHC) and shows reactivity with human, mouse, and rat samples, supporting comparative studies of pathway activation across these species. Phosphorylation at S256 drives 14-3-3 protein binding and nuclear export of FOXO1, inactivating its transcription of gluconeogenic, pro-apoptotic, and cell cycle arrest genes.

Investigators studying insulin resistance, type 2 diabetes, and tumor suppressor signaling use this reagent to monitor Akt pathway activity in cultured cell lysates and fixed tissue sections. The unconjugated format permits flexible pairing with labelled secondary antibodies and chromogenic, chemiluminescent, or fluorescence detection chemistries, and the polyclonal profile favors recognition of the phospho-S256 epitope even when flanking residues vary modestly. Short-term storage at 4 °C in the supplied buffer preserves activity, and aliquoting is recommended to limit freeze-thaw cycles.
