Raised in rabbit against a phosphorylated epitope, this polyclonal reagent from Antibodies.com selectively recognizes FKHR (also called FoxO1) when serine 256 is modified by Akt/PKB. Because Akt phosphorylation at Ser256 drives FoxO1 nuclear export and inactivation, the antibody serves as a direct read-out of PI3K/Akt pathway flux and is commonly used to follow insulin, IGF-1, and growth-factor signaling, along with downstream effects on hepatic gluconeogenesis, oxidative stress resistance, cell-cycle arrest, and apoptosis.

Validated applications include Western blot, immunohistochemistry, and immunofluorescence on human and mouse specimens. The antibody is supplied unconjugated as 100 µl at 1 mg/ml in phosphate buffered saline containing 0.05% sodium azide, pH approximately 7.2. As a polyclonal reagent, it offers broader epitope coverage that can be advantageous when monitoring dynamic phosphorylation states. Aliquoting is recommended to limit repeated freeze-thaw cycles of the stock solution.
