Raised in rabbit against the FKHR/FoxO1 transcription factor, this polyclonal reagent from Antibodies.com targets an epitope designated F313 within forkhead box protein O1. FoxO1 sits at a regulatory node in insulin and growth factor signaling, where Akt-mediated phosphorylation drives its nuclear export and inactivation, and it functions as a transcriptional activator of genes controlling gluconeogenesis, adipocyte differentiation, oxidative stress response, and apoptosis. The unconjugated IgG is supplied at 1 mg/ml in phosphate buffered saline containing 15 mM sodium azide at approximately pH 7.2, in a 50 µl vial that supports multiple Western blot, immunofluorescence, and immunohistochemistry experiments.

Validated on human, mouse, and rat samples, the antibody is appropriate for tracking endogenous FoxO1 in metabolic disease models of insulin resistance and type 2 diabetes, in skeletal muscle atrophy and autophagy studies, in hematopoietic and lymphocyte quiescence work, and in oncology where FoxO1 acts as a tumor suppressor in several tissue contexts. Because the reagent binds the native protein rather than an engineered tag, it can be applied to primary tissue lysates, cultured mammalian cell lines, and fixed or frozen sections prepared for staining or imaging.

As a polyclonal reagent, the antibody recognizes several epitopes on FoxO1, which can strengthen signal in applications where a single monoclonal may give weak detection. Sodium azide at 15 mM acts as a preservative, and aliquoting upon receipt is recommended to limit repeated freeze-thaw cycles; the buffered formulation at pH 7.2 is compatible with standard secondary antibody protocols including HRP- and fluorophore-conjugated reagents for chemiluminescence and fluorescence readouts.
