FHIT (Fragile Histidine Triad Diadenosine Triphosphatase) is a tumor suppressor encoded at the fragile site FRA3B on human chromosome 3p14.2, where it hydrolyzes diadenosine triphosphate and related diadenosine polyphosphates. Loss of FHIT expression is reported across a wide spectrum of epithelial cancers, including lung, head and neck, breast, and gastrointestinal tumors, so reagents that detect endogenous FHIT are commonly used in studies of tumor biology, fragile-site instability, and loss of heterozygosity. This unconjugated Rabbit polyclonal reagent from Antibodies.com enables detection of FHIT protein in samples from human, mouse, and rat, and has been validated for two routine applications: western blot analysis of cell and tissue lysates, and immunohistochemistry of formalin-fixed paraffin-embedded sections.

The antibody is supplied as 100 ul of a 1 mg/ml stock in PBS containing 0.1% sodium azide and 50% glycerol, a glycerol-rich buffer that tolerates refrigerated storage and occasional freeze-thaw cycles without appreciable loss of activity. The 100 ul pack size is convenient for pilot optimization of IHC antigen retrieval conditions, screening matched tumor and adjacent normal tissue pairs by WB, and follow-up validation of candidate FHIT-interacting partners by co-immunopilization. Because sodium azide is present as a preservative, the supplied buffer is not suitable for direct use in live-cell labeling or in assays sensitive to azide, and downstream workflows that require azide-free conditions should buffer-exchange the antibody before use.

Polyclonal reactivity means the reagent recognizes multiple epitopes on FHIT, which can improve signal on western blot when target expression is low and can be more forgiving across fixation conditions in IHC. As an unconjugated primary, it can be paired with HRP, alkaline phosphatase, biotin, or fluorescent secondary reagents chosen for the experimental read-out. Intended for research use only.
