The Antibodies.com FERMT3 polyclonal antibody is a rabbit-hosted IgG reagent raised against kindlin-3, the cytoskeletal adaptor encoded by FERMT3 that drives integrin activation in hematopoietic lineages. Supplied as an unconjugated antiserum in a 200 µl vial, it is validated for Western blot and immunofluorescence workflows on cultured cells and tissue sections, with reactivity confirmed against human samples and cross-reactivity demonstrated against mouse and rat orthologs.

FERMT3 (also called kindlin-3, URP2, or MIG2-like) is a FERM-domain focal adhesion protein expressed predominantly in leukocytes, platelets, and endothelial cells, where its binding to the cytoplasmic tail of β-integrins is required for inside-out activation of integrins such as αIIbβ3, αLβ2, and αMβ2. Loss-of-function mutations in FERMT3 cause Leukocyte Adhesion Deficiency type III (LAD-III), a primary immunodeficiency characterized by defective neutrophil adhesion, impaired platelet aggregation, and recurrent infections, making reliable FERMT3 detection a useful tool in adhesion, cytoskeleton, and immune-cell signaling research.

The polyclonal format allows recognition of several regions of the kindlin-3 molecule rather than a single linear epitope, which is helpful when probing full-length endogenous protein in primary leukocytes and megakaryocyte-derived cells. For Western blot, run reduced samples and detect under standard chemiluminescent conditions; for immunofluorescence, fixation in 4% paraformaldehyde is appropriate for both adherent and suspension hematopoietic cell lines. Refrigerate the vial and aliquot upon first use to limit freeze-thaw cycles and preserve antibody activity over the product shelf life.
