Raised in rabbit against the phosphorylated Y925 epitope of focal adhesion kinase (FAK/PTK2), this polyclonal antibody detects a key regulatory tyrosine that is phosphorylated by Src family kinases following integrin or growth factor stimulation. Phosphorylation at Y925 creates a Grb2 SH2 docking site, coupling FAK to Ras/MAPK signaling and linking focal adhesion turnover to downstream mitogenic pathways. Investigators studying cell migration, mechanotransduction, tumor invasion, and integrin-dependent signaling use this reagent to monitor pathway activation in cultured cells and tissue specimens.

Validated for western blot and immunohistochemistry, the antibody recognizes its target in human, mouse, and rat samples. It is supplied at 1 mg/ml in phosphate buffered saline (pH 7.2) containing 0.05% sodium azide, as a 100 µl vial of unconjugated IgG suitable for pairing with secondary antibodies. Aliquot upon receipt and store at -20°C; protect from light and avoid repeated freeze-thaw cycles to preserve phospho-specific reactivity. Working concentrations should be titrated empirically; common starting points fall around 1 µg/ml for western blot and 5-10 µg/ml for immunohistochemistry on paraffin sections, with optimization against antigen retrieval conditions.

This Antibodies.com reagent complements antibodies against total FAK and other phospho-sites such as Y397 and Y576/Y577, enabling parallel assessment of FAK activation state across experimental conditions.
