Raised in rabbit against Focal Adhesion Kinase (FAK, also known as PTK2), this unconjugated polyclonal antibody recognizes an epitope in the region surrounding residue 861 of the human protein. FAK is a non-receptor protein tyrosine kinase that localizes to focal adhesions and integrates signals from integrins, growth factor receptors, and mechanical cues to coordinate cell adhesion, spreading, migration, and survival. The Ab-861 region corresponds to a functionally important segment of the kinase domain, and phosphorylation at the neighboring Y861 site has been linked to FAK activation downstream of Src-family kinases and integrin engagement.

This reagent is validated for Western blot and immunohistochemistry on human samples and is supplied as 100 µl of unconjugated antiserum ready for downstream labeling with HRP, fluorophore, or biotin conjugates. For blotting applications, the antibody is commonly applied at 0.5–2 µg/ml following standard nitrocellulose or PVDF transfer, while immunohistochemistry working concentrations fall in the 1–10 µg/ml range depending on fixation and antigen retrieval conditions. Aliquot upon first use and store the vial at -20 °C in working volumes to limit freeze-thaw cycles; brief refrigeration of working aliquots is acceptable. Avoid sodium azide when downstream HRP conjugation is planned.

Researchers studying integrin signaling, focal adhesion turnover, tumor invasion, mechanotransduction, and cytoskeletal remodeling use this antibody to monitor endogenous FAK expression and modification status in cancer cell lines, primary fibroblasts, endothelial cells, and tissue sections. Distributed by Antibodies.com as a rabbit polyclonal reagent with documented human reactivity.
