Focal Adhesion Kinase (FAK), encoded by the PTK2 gene, is a ~125 kDa cytoplasmic non-receptor protein tyrosine kinase that localizes to focal adhesions and transduces signals from integrin engagement and growth factor receptors into cytoskeletal reorganization, cell migration, proliferation, and survival responses. Dysregulation of FAK is widely documented across many solid tumour types as well as in fibrotic and angiogenic processes, making it a heavily studied target in cell biology and oncology research.
This Antibodies.com reagent is a rabbit polyclonal antibody raised against a region of human FAK centred near residue 843, a site associated with regulatory phosphorylation that modulates catalytic output and SH2-domain partner binding. Because polyclonal reagents recognize multiple epitopes within the immunogen, the antibody provides robust detection of native and denatured FAK across a range of downstream applications.
The antibody is supplied as 100 µl of unconjugated antiserum in a single vial, allowing the end user to pair it with HRP-, biotin-, or fluorophore-conjugated secondary reagents according to experimental design. Validation has been performed for Western blot detection of the ~125 kDa full-length FAK band in cell lysates and for immunohistochemistry localization on formalin-fixed, paraffin-embedded tissue sections. Reactivity is confirmed for human samples; cross-reactivity with mouse, rat, or other species has not been established by the manufacturer.
Working dilution guidance commonly falls between 1:500 and 1:2000 for Western blot and 1:50 to 1:200 for immunohistochemistry, though individual titration is recommended for each sample type. Store the vial frozen at -20 °C in working aliquots to limit freeze-thaw cycles, and protect from light when using fluorescent secondary detection. The product is intended for research use only and supports studies of integrin signalling, focal adhesion biology, tumour invasion, and mechanotransduction.
