This unconjugated rabbit polyclonal reagent from Antibodies.com is raised against the Eyes-Absent (EYA) family proteins EYA1 and EYA4 and is supplied as 100 µl of a 1 mg/ml stock in phosphate buffered saline (PBS) at approximately pH 7.2, with 15 mM sodium azide included as a microbial preservative. The polyclonal format means the antiserum recognizes multiple epitopes on each target, which is helpful when probing paralogs that differ by tissue-specific isoforms or post-translational modifications that might mask a single linear site. The reagent is validated for Western blot and shows reactivity with human and rat samples, supporting translational work that moves between rodent disease models and patient material.

EYA1 is the gene most strongly linked to branchio-oto-renal (BOR) syndrome, in which heterozygous loss-of-function variants disrupt the development of the branchial arches, the inner ear, and the kidney, while dominant EYA4 variants are better known for sensorineural hearing loss and familial dilated cardiomyopathy. At the molecular level, both proteins function as transcriptional coactivators that partner with SIX1–SIX6 homeodomain factors and carry intrinsic protein phosphatase activity; EYA1 in particular dephosphorylates H2AX at tyrosine 142 during the DNA damage checkpoint, a reaction that helps determine whether cells proceed toward repair or apoptosis after genotoxic stress. Researchers apply anti-EYA1/EYA4 antibodies to Western blot lysates from developing organs, hypertrophic cardiomyocyte assays, and DNA damage response experiments.

For Western blot, 20–40 µg of total protein per lane is a practical starting point, resolved on standard SDS-PAGE and transferred to a suitable membrane before incubation with the primary antibody at empirically optimized dilutions. Because the antibody is unconjugated, users can pair it with their preferred HRP- or fluorophore-conjugated secondary antibody. Short-term storage at 4 °C is appropriate for routine use; for longer intervals the vial should be aliquoted and frozen to limit freeze-thaw cycles, and the reagent should be protected from light to preserve the sodium azide-stabilized formulation.
