ERK1 and ERK2 (also known as MAPK3 and MAPK1) are serine/threonine kinases that sit at the core of the Ras-Raf-MEK-ERK signal transduction cascade, transmitting mitogenic signals from cell surface receptors to the nucleus. Phosphorylation at the T-E-Y motif drives activation and downstream effects on cell proliferation, differentiation, and survival, which is why ERK1/2 is among the most heavily studied targets in oncology, neurobiology, and inflammation research.

This unconjugated rabbit polyclonal reagent from Antibodies.com was raised against a synthetic peptide centred on the L352 region of ERK1/2, a site commonly used to track total kinase levels regardless of activation state. The antibody is supplied as 100 µL of a 1 mg/mL stock in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2, and is validated for western blot and immunohistochemistry on human, mouse, and rat samples.

For storage, aliquot upon receipt and freeze at -20 °C in working volumes to limit freeze-thaw cycles; the sodium azide preservative makes the formulation unsuitable for live-cell work. The polyclonal nature of the reagent gives broader epitope coverage than a monoclonal, which is helpful when probing for ERK1/2 across multiple species or detecting both isoforms simultaneously.
