This polyclonal antibody from Antibodies.com targets ERCC3, the ATP-dependent DNA helicase also known as XPB. ERCC3 is a core subunit of the TFIIH transcription-repair complex and unwinds DNA in the 5' to 3' direction, an activity required for nucleotide excision repair and for RNA polymerase II transcription initiation. The reagent is raised in rabbit and supplied as 100 µl of unconjugated antiserum for use in Western blotting and immunohistochemistry on human, mouse and rat samples. In research settings, the antibody supports studies of UV-induced DNA damage response, TFIIH assembly, and the molecular basis of nucleotide excision repair disorders such as xeroderma pigmentosum, Cockayne syndrome and trichothiodystrophy, where inherited ERCC3 mutations compromise repair capacity. The unconjugated format allows free pairing with HRP-, biotin- or fluorophore-conjugated secondaries, fitting chromogenic Western blot development as well as immunoperoxidase or fluorescence tissue staining. Endogenous ERCC3 expression can therefore be assessed in cell lysates and in fixed tissue sections using standard immunoblotting and immunostaining protocols.
