Phospho-specific detection of activated ERBB2 (HER2)

This unconjugated rabbit polyclonal IgG antibody from Antibodies.com selectively recognizes human ERBB2 only when residue tyrosine 1248 in the C-terminal regulatory tail is phosphorylated. The Y1248 site is a major autophosphorylation target that reports active receptor signaling downstream of HER2 heterodimerization with other EGFR family members, making this reagent appropriate for studies of receptor activation dynamics rather than total HER2 protein levels.

Validated for Western blot on human samples, the antibody is supplied as 200 µl of liquid in a vial. The polyclonal format provides recognition of multiple epitopes surrounding the phospho-Y1248 motif, which can strengthen signal in WB where a single monoclonal may fail to detect the modified receptor under reduced antigen conditions. Researchers commonly pair this reagent with a total-ERBB2 loading control to normalize activated-to-total receptor ratios in cell line and tumor lysates, and to probe pathway inhibition by small-molecule HER2 kinase inhibitors such as lapatinib or neratinib in HER2-driven breast and gastric cancer models.

Storage recommendations: aliquot upon receipt to limit freeze-thaw cycles, and store at -20 °C in a manual defrost freezer. The unconjugated IgG format pairs with anti-rabbit HRP or fluorescent secondary antibodies depending on the chosen Western blot detection chemistry. For research use only; not intended for diagnostic or therapeutic applications.
