Affinity-purified rabbit polyclonal IgG, raised against Ephrin type-B receptor 3 (EPHB3) and supplied as an unconjugated 200 µl vial for Western blot immunoblotting. Produced by Antibodies.com using a synthetic peptide immunogen derived from the human EPHB3 sequence, the antibody is purified by epitope-specific affinity chromatography to greater than 95% purity by SDS-PAGE and validated for detection of endogenous EPHB3 protein in human, mouse, and rat samples.
EPHB3 is a transmembrane receptor tyrosine kinase of the EphB family that transduces contact-dependent forward signals from membrane-anchored ephrin-B ligands on adjacent cells. The receptor is well documented as a positional cue in the intestinal epithelium, where Wnt-driven EphB3 expression guides Paneth cells to the crypt base and maintains the proliferative progenitor compartment, and in the developing central nervous system, where EphB/ephrin-B signaling regulates axon guidance and midline crossing in the spinal cord.
This polyclonal format recognizes multiple epitopes on EPHB3, which strengthens band signal in transfected overexpression systems as well as in samples with low endogenous expression, and the unconjugated delivery leaves the choice of secondary reporter open to the laboratory, allowing HRP-based chemiluminescent, fluorophore-based, or biotin–streptavidin detection on the same primary blot.
For routine handling, aliquot the antibody upon first use to limit repeated freeze-thaw cycles and refrigerate the working stock between applications. The reagent pairs naturally with standard loading controls such as anti-GAPDH or anti-β-actin when normalizing EPHB3 signal across treatment conditions, and is supplied for research use only, supporting studies of axon guidance, intestinal crypt regeneration, and the documented role of EPHB3 as a tumor and invasion suppressor that is silenced at the adenoma–carcinoma transition in colorectal cancer.
