A rabbit polyclonal antibody raised against eukaryotic translation initiation factor 4E, supplied by Antibodies.com as an unconjugated reagent in a 100 µl volume. eIF4E is the cap-binding subunit of the eIF4F translation initiation complex and recognizes the 7-methylguanosine cap at the 5′ end of cellular mRNAs. The protein runs near 25 kDa on reducing SDS-PAGE gels and is phosphorylated on Ser209 by the Mnk1 and Mnk2 kinases, a modification commonly monitored as a downstream readout of MAPK pathway activation.

This polyclonal reagent was validated for Western blot detection of endogenous eIF4E in lysates prepared from human, mouse, rat, and monkey origin cell lines and primary tissues. It is also suited to immunohistochemistry on formalin-fixed paraffin-embedded sections, where staining is observed in the cytoplasm and, in many tumor specimens, in the nuclear compartment. In addition, the antibody has been used as a capture reagent in indirect ELISA setups for relative quantitation of eIF4E in cell extract samples.

For storage, the antibody is shipped refrigerated and should be aliquoted on first use to limit repeated freeze-thaw cycles. Short-term storage at 4°C is appropriate for working aliquots, while the stock should be kept at -20°C for longer-term stability. The reagent is intended for research use only and is not approved for diagnostic applications.
