The Antibodies.com Anti-eIF2α (2E4) antibody is a mouse monoclonal reagent that recognizes the alpha subunit of eukaryotic translation initiation factor 2, a roughly 36 kDa regulatory protein that gates the initiation of protein synthesis. The unconjugated IgG is supplied as a 100 µL liquid aliquot in 0.1 M Tris-Glycine buffer (pH 7.4) with 150 mM NaCl, 50% glycerol, and 0.2% sodium azide as a preservative, and it is validated for Western blot and immunofluorescence applications on human, mouse, and rat samples.

Researchers use the 2E4 clone to monitor the integrated stress response, in which the kinases PERK, GCN2, HRI, and PKR phosphorylate eIF2α at Ser51 to repress global cap-dependent translation while selectively permitting translation of ATF4-driven stress-response transcripts. The antibody is therefore widely deployed in studies of the unfolded protein response in ER-stressed cells, viral infection models that activate PKR, amino acid deprivation and leucine-starvation assays, redox and heme-deficiency protocols, and investigations of ischemic injury, neurodegeneration, metabolic dysfunction, and tumor cell survival under hypoxic or nutrient-limited conditions. The 2E4 epitope is also frequently cited in work on chemotherapy-induced stress signaling and in neuronal damage models examining synaptic dysfunction after prolonged stress.
