This rabbit polyclonal antibody from Antibodies.com targets EED (Embryonic Ectoderm Development), a core scaffold subunit of Polycomb Repressive Complex 2 (PRC2). PRC2 catalyzes trimethylation of histone H3 at lysine 27 (H3K27me3), a repressive chromatin modification central to X-chromosome inactivation, genomic imprinting, stem-cell pluripotency, and lineage commitment. EED contains seven WD40-repeat domains that form a beta-propeller binding pocket for H3K27me3, and this recognition allosterically activates the methyltransferase subunit EZH2 to propagate Polycomb silencing across cell division.

The unconjugated reagent is supplied as 100 microlitres of polyclonal antiserum raised in rabbit, validated for Western blot, immunohistochemistry, and immunofluorescence on human samples. In Western blot of whole-cell lysates, the antibody detects an endogenous band at approximately 50 kDa, in line with the predicted molecular weight of full-length human EED. By immunofluorescence on fixed cells, staining is predominantly nuclear, consistent with the chromatin-associated function of PRC2. On formalin-fixed paraffin-embedded tissue sections, immunohistochemistry reveals nuclear labeling in cells retaining active Polycomb-mediated repression.

Storage recommendations include keeping the vial refrigerated, protecting it from light, and aliquoting into working volumes to limit freeze-thaw cycles if frozen for longer-term storage. The reagent is intended for research use only and is not validated for diagnostic or therapeutic procedures. Investigators working in cancer epigenetics, developmental biology, and chromatin research will find it useful for monitoring PRC2 complex integrity and H3K27me3 pathway status in cell-based and tissue-based models.
