Raised in rabbit and supplied by Antibodies.com as an unconjugated polyclonal reagent, this anti-EDEM1 immunoglobulin is presented at 1 mg/ml in phosphate-buffered saline containing 15 mM sodium azide at approximately pH 7.2, and is provided as 100 µl of liquid in a single-use vial. EDEM1 (ER degradation-enhancing alpha-mannosidase-like protein 1) operates at the front end of the ER-associated degradation (ERAD) pathway, recognizing N-glycosylated, misfolded client proteins within the endoplasmic reticulum lumen and supporting their retrotranslocation and proteasomal disposal. It functions as an alpha-1,2-mannosidase that trims Man9GlcNAc2 glycans on misfolded conformers to a Man8GlcNAc2 mark, a signal subsequently read by the lectins OS-9 and XTP3-B to deliver substrates to the SEL1L-HRD1 retrotranslocon complex. Disease-relevant ERAD clients that EDEM1 helps to clear include the Z variant of alpha-1-antitrypsin and the most common cystic fibrosis mutant of CFTR (ΔF508). The reagent is documented as reactive against the human, mouse, and rat orthologs, supporting comparative work across disease models that interrogate hepatocellular stress, neurodegeneration, and tumor cell adaptation to proteasome inhibition or tunicamycin-induced unfolded protein response activation. Validated for western blot detection under denaturing, reducing SDS-PAGE conditions, the antibody yields the expected EDEM1 band at the predicted molecular weight, with chemiluminescent read-out typically giving sharp signal at low background. Researchers commonly use it alongside classical ER stress markers such as BiP/GRP78, CHOP, and XBP1s, or in pulse-chase assays that follow the half-life of model ERAD substrates to assess EDEM1 activity. The sodium azide-containing buffer keeps the reagent stable when stored refrigerated between uses; aliquoting is recommended to limit freeze-thaw cycles if the vial is held for extended periods.
