Rabbit polyclonal antibodies raised against dihydropyrimidinase (DPYS) are available from Antibodies.com as a 100 µL vial at 1 mg/ml, formulated in PBS with 0.1% sodium azide and 50% glycerol. The unconjugated reagent targets the cytosolic enzyme that catalyzes the second step of pyrimidine catabolism, converting 5,6-dihydrouracil and 5,6-dihydrothymine to the corresponding N-carbamyl-β-amino acids, and is validated for Western blot and immunohistochemistry on human, mouse, and rat samples.

DPYS is most abundantly expressed in liver and kidney, where pyrimidine breakdown is most active, and the protein migrates near its expected ~56 kDa molecular weight in reducing SDS-PAGE, giving a clean band in Western blot when adequate lysis conditions are used. For IHC, heat-induced antigen retrieval with citrate buffer (pH 6.0) is commonly applied to formalin-fixed paraffin-embedded sections to expose the epitope in tissue slices.

Store the glycerol-stabilized stock at -20°C in working aliquots to limit freeze-thaw cycles; short-term handling at 4°C is fine. The reagent is for research use only, and supports studies of pyrimidine metabolism, fluoropyrimidine drug toxicity (DPYS activity modulates 5-FU catabolism), and the rare metabolic disorder dihydropyrimidinuria associated with DPYS mutations.
