The polyclonal reagent from Antibodies.com is raised in rabbit and selectively recognizes DOK1 only when phosphorylated at the Y398 residue, a regulatory site modified by Src-family and receptor tyrosine kinases. DOK1 is a 62 kDa adaptor that recruits RasGAP and Nck to attenuate Ras-MAPK and PI3K-Akt signaling downstream of immune receptors, oncogenic kinases, and growth factor stimulation; phosphorylation at Y398 alters SH2-mediated partner binding and is widely used as a readout of pathway activation.

Supplied as 100 µL of unconjugated IgG, the antibody is validated for Western blot and immunohistochemistry across human, mouse, and rat samples. Investigators apply it to study negative-feedback signaling downstream of B-cell and T-cell receptors, to compare DOK1 activity in leukemia or solid-tumor panels against matched normal tissue, and to follow modulation of adaptor phosphorylation in cytokine-stimulated immune-cell cultures.

For best results, lysates are prepared with standard phosphatase inhibitors to preserve the phospho-epitope, and immunohistochemistry is performed on formalin-fixed paraffin-embedded sections following antigen retrieval. Because the reagent is phosphorylation-state specific, prior lambda-phosphatase treatment of samples abolishes signal, providing a convenient internal specificity control.
