The DOK1 (Docking protein 1) polyclonal reagent from Antibodies.com is an unconjugated rabbit IgG primary antibody intended for western blot detection of the endogenous protein in human, mouse, and rat samples.

DOK1 is a 62 kDa cytosolic adapter assembled from an N-terminal pleckstrin homology (PH) domain, a central phosphotyrosine-binding (PTB) domain, and a C-terminal tail carrying multiple tyrosine residues. Upon receptor tyrosine kinase, B-cell receptor, and integrin engagement, these tyrosines become phosphorylated and recruit SH2-bearing partners such as RasGAP, Nck, and CrkL, allowing DOK1 to dampen Ras/MAPK and PI3K/Akt signaling. It functions as a feedback inhibitor in hematopoietic cells and modulates proliferation, adhesion, and cytoskeletal remodeling downstream of growth factor and immune receptors.

Each vial contains 200 µl of affinity-purified antibody, suitable for probing whole-cell lysates, tumor-derived lines, and tissue homogenates by western blot. Short-term storage at 4 °C preserves activity for routine use; aliquot and freeze for long-term retention.

Typical applications include immunoblotting of phospho-DOK1 in stimulated immune cells, mapping of insulin- and growth-factor-driven pathways where DOK1 acts as a negative regulator, and characterization of B-cell and T-cell receptor signaling in primary lymphocytes and rodent models.
