Raised in rabbit and affinity-purified as a polyclonal IgG, this Antibodies.com primary antibody targets DOK1 (Docking Protein 1), a 52 kDa cytoplasmic adapter that operates downstream of multiple receptor and non-receptor tyrosine kinases. The unconjugated reagent is validated for western blot detection of endogenous DOK1 in human, mouse, and rat samples, and is supplied as 100 µl per vial for direct pairing with HRP-, biotin-, or fluorophore-labeled secondary antibodies.

For immunoblotting, researchers typically resolve whole-cell lysates prepared under standard RIPA conditions by SDS-PAGE, transfer to nitrocellulose or PVDF membranes, and titrate the primary empirically; working dilutions reported for comparable anti-DOK1 polyclonal reagents fall around 1:500 to 1:2,000. The reagent should be stored at -20 °C in working aliquots to limit repeated freeze-thaw cycles and preserve activity across multiple experiments, and each vial ships as a liquid formulation ready for dilution into blocking buffer.

DOK1 carries an N-terminal pleckstrin homology domain, a C-terminal phosphotyrosine-binding domain, and ten PXXP SH3 recognition motifs that mediate assembly of downstream signaling complexes. It serves as a major substrate for c-Kit, EGF receptor, PDGF receptor, insulin receptor, v-abl, and vFPS kinases; once phosphorylated, DOK1 forms a complex with Ras GAP and feeds back negatively on the Ras/Raf/MAPK cascade, contributing to inhibition of cell proliferation and transformation, as well as control of cell spreading and migration. The adapter is also implicated in T-cell and B-cell antigen receptor pathways, making this reagent useful for research into hematopoietic receptor feedback, insulin pathway regulation, and downstream signaling of receptor tyrosine kinases.
