Phosphorylation of the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) at tyrosine 2609 marks the activated form of this kinase that orchestrates non-homologous end joining repair of DNA double-strand breaks. This rabbit polyclonal antibody from Antibodies.com is raised against the phospho-Y2609 epitope and is validated for immunohistochemistry on human tissue, supporting studies of radiation-induced DNA damage, genomic instability, and tumor radiosensitivity in translational oncology research.

The reagent is supplied as an unconjugated, affinity-purified IgG at 1 mg/ml in phosphate buffered saline containing 0.05% sodium azide at approximately pH 7.2. Each vial contains 100 µl of working stock. For short-term use the vial can be refrigerated; for long-term preservation of phospho-specific binding activity, aliquot and freeze at -20°C, protect from light, and limit freeze-thaw cycles.

The recommended IHC workflow includes deparaffinization, heat-induced antigen retrieval in citrate buffer at pH 6.0, blocking in serum, and detection with an HRP-conjugated goat anti-rabbit secondary antibody followed by DAB chromogen and hematoxylin counterstain. The reagent is reactive with human DNA-PKcs phosphorylated at tyrosine 2609 and is intended for research use only in studies of DNA damage signaling and repair.
