The DDDDK tag (also written as the FLAG DYKDDDDK epitope) is an 8-residue hydrophilic peptide fused to recombinant proteins for affinity purification and tracking. This unconjugated mouse polyclonal IgG reagent recognizes the DDDDK epitope and detects FLAG-tagged fusion proteins across standard immunology workflows.

From Antibodies.com, the antibody is supplied at 50 µl per vial and is validated for Western blot, immunohistochemistry, immunofluorescence, and immunoprecipitation. Polyclonal clonality means multiple epitopes on the DDDDK sequence are recognized, which can strengthen signal on weakly expressed tagged constructs where single-clone binders may give marginal detection.

The small size of the DDDDK tag makes it unlikely to disrupt folding or function of the host protein, so the antibody is commonly applied to confirm expression of tagged constructs during purification, to localize fusion proteins in fixed cells and tissue sections, and to pull down tagged interactors. Because the host is mouse, anti-mouse IgG secondary reagents are used for downstream visualization.

For handling, store refrigerated for short-term use and freeze the antibody in small working aliquots to limit repeated freeze-thaw cycles, which can degrade antibody activity over time.
