Targeting one of the most clinically significant drug-metabolizing enzymes in the human body, this polyclonal reagent recognizes Cytochrome P450 3A4 (CYP3A4), the dominant CYP isoform expressed in adult liver and intestinal epithelium that oxidatively metabolizes roughly half of all marketed pharmaceuticals.

From Antibodies.com, the antibody is supplied as 100 µl of unconjugated antiserum raised in rabbit and is validated for Western blot and immunohistochemistry workflows. Because the target protein governs the clearance of a large share of clinically prescribed drugs, the reagent is commonly used in translational studies of drug-drug interactions, induction and inhibition kinetics, inter-individual variability in hepatic metabolism, and altered CYP3A4 expression in conditions such as cirrhosis, non-alcoholic fatty liver disease, and several solid tumors where CYP3A4 levels are frequently dysregulated.

In Western blot, the antibody is expected to detect the characteristic ~57 kDa band corresponding to CYP3A4 in liver microsomal fractions and whole-cell lysates. For immunohistochemistry, it can be applied to formalin-fixed, paraffin-embedded tissue sections to visualize CYP3A4 distribution within hepatocytes and enterocytes of the small intestine. Researchers should aliquot the vial upon receipt to limit freeze-thaw cycles of the antiserum and store it refrigerated for short-term use or frozen for longer-term retention.
