Raised in rabbit against human CYB5R3 (NADH-cytochrome b5 reductase 3), this unconjugated polyclonal antibody from Antibodies.com is supplied as 100 µl of ready-to-use reagent and is validated for Western blot and immunohistochemistry on human samples. CYB5R3 is the FAD-containing enzyme that reduces methemoglobin back to functional hemoglobin inside erythrocytes, and in its membrane-anchored isoform it also contributes to fatty-acid elongation, cholesterol biosynthesis, and cytochrome P450-mediated drug metabolism across the endoplasmic reticulum and outer mitochondrial membrane.

Researchers investigating recessive congenital methemoglobinemia, erythrocyte redox balance, or hepatic xenobiotic handling use this reagent to detect both the soluble erythrocyte form (~30 kDa) and the membrane-bound form (~34 kDa) of the reductase. The polyclonal format allows recognition of multiple epitopes on the target, which is helpful when probing tissue sections where epitope accessibility varies, such as liver, kidney, skeletal muscle, and cardiac tissue. For Western blot, lysates prepared in standard RIPA buffer with freshly added protease inhibitors give clean signals, and the antibody should be aliquoted on arrival and stored refrigerated; repeated freeze-thaw cycles will reduce the working titer.

Because the antibody is supplied unconjugated, it pairs with the secondary detection system of the researcher's choice, including HRP-, biotin-, or fluorophore-conjugated anti-rabbit IgG reagents. The human reactivity profile makes it directly relevant for translational work linking CYB5R3 expression to methemoglobin levels, lipid-handling disorders, and P450-dependent drug clearance in patient-derived specimens.
