Phosphorylation-state-specific readout for canonical Wnt signalling: this rabbit polyclonal IgG reagent from Antibodies.com recognises β-catenin (CTNNB1) only when serine 37 within the N-terminal phosphodegron is phosphorylated. The S37 site, together with the adjacent S33 and T41 residues, forms the canonical β-TrCP/SCF recognition motif that flags cytosolic β-catenin for ubiquitination and proteasomal destruction, so pS37 signal tracks ligand-driven pathway activation inversely: high phosphodegron phosphorylation corresponds to an active destruction complex, while loss of the pS37 band reflects Wnt-induced stabilisation of the transcriptional co-activator pool.

The antiserum is supplied unconjugated as 200 µl aliquots and has been validated for Western blot and immunohistochemistry on human, mouse, and rat tissue, covering the model systems most commonly used in colorectal and hepatocellular carcinoma research, intestinal stem-cell biology, and developmental signalling studies. Investigators comparing Wnt ligand stimulation, GSK3β inhibition, or upstream kinase perturbations use the reagent to monitor changes in phosphodegron stoichiometry across treatment arms. Aliquot upon receipt and store at -20 °C frozen; repeated freeze-thaw cycles erode phospho-epitope binding activity over time.
