Cyclic AMP-responsive element-binding protein 3 (CREB3) is an endoplasmic-reticulum-membrane transcription factor of the ATF/CREB family that is cleaved during the unfolded protein response, releasing an N-terminal fragment that enters the nucleus to regulate ER-stress and lipid-metabolism genes. Endogenous CREB3 can be visualised by IF in fixed cells to follow nuclear translocation after ER stress, while Western blot detects both the membrane-bound precursor and the processed nuclear fragment. The unconjugated format lets the user pair it with a secondary detection reagent of choice, such as HRP for chemiluminescent Western blot or fluorophore-conjugated secondaries for IF.
