Produced in rabbit and supplied as 100 µl of unconjugated antiserum, this Antibodies.com primary antibody targets the WD-repeat actin-binding protein Coronin 1B, a coronin family member involved in cytoskeletal remodeling. Validated for Western blot and immunohistochemistry workflows on human samples, the reagent demonstrates species specificity for the human antigen.
Coronin 1B accumulates at the leading edge of motile cells, where it modulates Arp2/3-mediated actin branching, stabilizes lamellipodial protrusions, and promotes turnover of cofilin-associated actin filaments. Published studies have linked its activity to invadopodia formation in carcinoma cells, chemotactic migration in fibroblasts and leukocytes, and coordination of N-WASP signaling downstream of Rac1 activation.
Researchers investigating cell motility, tumor invasion, phagocytic cup dynamics, receptor endocytosis, or neuronal pathfinding use this reagent to probe endogenous Coronin 1B distribution in fixed tissue sections and total cell lysates. The polyclonal host response yields broad epitope coverage across the human antigen, supporting detection of full-length and processed forms in WB membranes and IHC sections.
For Western blot, antibody dilutions in standard Tris-buffered saline with non-fat milk or BSA blocking are typically used; for IHC, antigen retrieval with citrate buffer followed by HRP or fluorescence-based secondary detection provides clean cytoplasmic and membrane-ruffle staining patterns consistent with the protein's known subcellular localization.
Store the vial refrigerated; avoid repeated freeze-thaw cycles. Aliquot upon first use if long-term storage is planned. The unconjugated format integrates readily with custom biotin, HRP, or fluorophore labeling chemistries for multiplexed imaging or signal amplification in research use only applications.
