Raised in rabbits against a defined epitope surrounding lysine 257, this polyclonal reagent targets the gap junction protein Connexin 43 (GJA1), a transmembrane connexin that assembles into hexameric hemichannels and docks between adjacent cells to form gap junction intercellular communication channels. Distributed across cardiomyocytes, astrocytes, osteoblasts, and epithelial layers, Cx43 is studied in the context of cardiac electrical coupling, astrocyte–neuron signaling, wound re-epithelialization, and tumor growth control, where loss or mis-localization of the protein alters tissue-level coordination. The preparation is unconjugated, and the product is supplied at 1 mg/ml in phosphate buffered saline with 0.05% sodium azide at approximately pH 7.2, packaged as a 100 µl vial. Cross-reactivity has been validated on human and mouse samples, supporting routine use in basic and translational research, with Western blot, immunohistochemistry, and immunofluorescence listed as recommended applications by Antibodies.com. Aliquot on first use to limit freeze-thaw cycles, keep working dilutions refrigerated, and store stock frozen at -20 °C for long-term retention of titer. The K257 region falls within the cytoplasmic C-terminal domain of Connexin 43, a segment enriched in regulatory serine and tyrosine phosphorylation sites that gate channel opening, making this reagent a practical choice for work tracking modification-dependent trafficking of Cx43 to and from the plasma membrane.
