This rabbit polyclonal antibody from Antibodies.com is raised against COL10A1, the gene encoding the alpha-1 chain of type X collagen, a short-chain collagen expressed almost exclusively by hypertrophic chondrocytes within the growth plate cartilage. The reagent is supplied as 100 µl of unconjugated antiserum in a single-use vial format and is validated for western blot (WB) and immunohistochemistry (IHC) applications on human tissue and cell samples.

Type X collagen is a well-established molecular marker of chondrocyte hypertrophy and the terminal differentiation programme that precedes endochondral ossification, making this reagent valuable for studies of skeletal development, fracture callus remodelling, osteoarthritis progression, and Schmid metaphyseal chondrodysplasia (SMCD), a skeletal dysplasia caused by dominant-negative COL10A1 mutations. Because the target is restricted to the hypertrophic zone of cartilage, the antibody is widely used to map zonal architecture in growth plate sections and to monitor COL10A1 induction during in vitro chondrogenic differentiation of mesenchymal progenitor or stem cell populations. The polyclonal format also increases the likelihood of recognising multiple epitopes on the alpha-1 chain, which is helpful when probing processed or partially degraded fragments in cartilage extracts.

For storage, refrigerate the vial upon arrival and aliquot into smaller volumes to limit freeze-thaw cycles during long-term storage. The unconjugated format allows flexible pairing with HRP-, biotin-, or fluorophore-conjugated secondary reagents selected by the end user, supporting both chromogenic and fluorescence-based detection in WB membrane and IHC section workflows.
