Description

This Antibodies.com reagent is a rabbit polyclonal antibody that recognises Cofilin1/2 (T88) and is provided at 1 mg/ml in phosphate buffered saline (PBS) with 0.05% sodium azide, at approximately pH 7.2. The unconjugated antibody has been validated for Western blot (WB) and immunocytochemistry/immunofluorescence (ICC/IF), and it reacts with human, mouse and rat samples.

It was raised against a synthetic peptide corresponding to amino acids 55–100 of human Cofilin1, and the affinity-purified antibody specifically detects endogenous levels of Cofilin1/2 protein. The reagent was isolated from rabbit antiserum by affinity chromatography using the epitope-specific immunogen, with a purity of greater than 95% as determined by SDS-PAGE. The target protein migrates at approximately 19 kDa.

Cofilin is ubiquitously expressed in eukaryotic cells, where it binds actin and regulates the rapid cycling of actin assembly and disassembly essential for cellular viability. As a low-molecular-weight protein, it binds filamentous F-actin by bridging two longitudinally associated actin subunits, changing the F-actin filament twist. This activity is enabled by dephosphorylation of cofilin at Ser 3, for example by factors such as opsonized zymosan, whereas LIM kinase 1, a serine kinase, phosphorylates cofilin and renders it unable to bind and depolymerise F-actin.
