Produced in rabbit as a polyclonal reagent, this unconjugated primary antibody from Antibodies.com selectively recognizes the cleaved (activated) form of caspase-3, a cysteine protease that acts as a central executioner of programmed cell death. The antibody binds the p17 fragment generated during apoptotic processing, allowing researchers to monitor caspase-3 cleavage as a hallmark indicator of apoptotic signaling cascades in basic research.

Validated for Western blot and immunohistochemistry workflows, the reagent reacts with Human, Mouse, and Rat samples, supporting cross-species studies of apoptosis in cell culture systems, fixed tissue sections, and whole-cell lysate preparations. In Western blot, the cleaved fragment is typically detected at approximately 17 kDa, with the full-length pro-caspase-3 zymogen near 35 kDa serving as an internal control for activation efficiency. The unconjugated format permits flexible pairing with HRP-conjugated, biotinylated, or fluorophore-conjugated secondary antibodies selected to match individual experimental designs.

Each vial is supplied as 100 µl of liquid antibody, providing sufficient volume for multiple blotting and staining runs. For research use only, the reagent is best stored at -20 °C in single-use aliquots to preserve binding activity across repeated freeze-thaw cycles. Recommended working dilutions generally fall between 1:500 and 1:2000 for Western blot applications, and between 1:100 and 1:500 for immunohistochemistry on formalin-fixed paraffin-embedded sections, depending on tissue type, antigen density, and detection chemistry.
