A rabbit polyclonal IgG reagent from Antibodies.com is raised against the muscle nicotinic acetylcholine receptor beta1 subunit (CHRNB1), a pentameric ligand-gated cation channel encoded on chromosome 17p13.1. The antiserum is produced against a synthetic peptide corresponding to human CHRNB1 amino acids 41–90, with a predicted subunit molecular weight near 56.7 kDa on denaturing gels. Supplied as 100 µL of unconjugated reagent in liquid form, the antibody is recommended for western blot at 1:500–1:2000 dilution and recognizes endogenous CHRNB1 in human and mouse samples. The unconjugated format allows users to pair it with anti-rabbit HRP, biotin, or fluorescent secondaries for chemiluminescent or multiplex detection schemes.

CHRNB1 assembles with two α1 subunits plus a δ subunit and either a γ subunit (embryonic AChR) or an ε subunit (adult AChR) to form the motor endplate receptor that drives neuromuscular junction transmission. Dominant gain-of-function missense mutations in the M2 pore-lining helix, including V266M and L263M, slow channel closure, prolong endplate currents, and produce slow-channel congenital myasthenic syndrome type 2A (CMS2A, OMIM 616313) with endplate myopathy, calcium overload, and progressive axial and limb weakness. Recessive CHRNB1 mutations are similarly linked to congenital myasthenic syndrome with acetylcholine receptor deficiency (CMS-ACHRD), and phosphorylation of the β1 cytoplasmic loop at residue βY390 is a known regulatory mark of receptor clustering at the synapse.

Validated research applications include confirming subunit expression in skeletal muscle and myotube lysates, screening patient-derived samples for receptor assembly defects linked to CMS, profiling autoantibody reactivity in myasthenia gravis workflows, and following agrin-induced AChR stabilization in cultured muscle cells. For storage, aliquot the antiserum upon arrival and keep refrigerated; limiting freeze-thaw cycles helps preserve titre across months of routine western blot use.
